چکیده

Purpose Active cellular therapy (ACT) using ex-vivo expanded T cells from patients with cancer, obtained by apheresis, can represent a viable source for anti-cancer directed cellular therapy. We established a T cell expansion protocol using 2 rounds of re-stimulation with TAA peptides along with IL-2, IL-15 and IL-21. In order to gauge the ex-vivo cellular reactivity as well as the potential to successful expand antigen-specific T cells from patients with pancreatic cancer, we established a screening assay using whole-heparin blood, to gauge for TAA reactivity (NY-ESO-1, survivin and mesothelin) and control antigens (EBNA-1, EBNA-3, CMVpp65).

Evaluation of in vitro Effect of Pyocyanine Pigment on Interleukin-2 Secretion from Peripheral Blood Mononuclear Cells in Cancer Patients

Background and Objective: The pyocyanine pigment in Pseudomonas aeruginosa stimulates blood cells to secrete IL-2. IL-2 cytokine is an activator of cytotoxic T cells and natural killer cells. These cells destroy the target cells of patients with cancer. This study aimed to evaluate the effects of pyocyanine on the IL-2 secretion from peripheral blood mononuclear cells (PBMCs) in patients with b...

Different Expressions of Specific Transcription Factors of Th1 (T-bet) and Th2 cells (GATA-3) by Peripheral Blood Mononuclear Cells From Patients With Multiple Sclerosis

Introduction: Multiple Sclerosis (MS) is an inflammatory disorder caused by self-reactive Th1 lymphocytes, while Th2 cells may confer protection. The Th1 and Th2 cell differentiation are regulated by specific transcription factors, especially T-bet and GATA-3, respectively. This investigation aimed to measure the T-bet and GATA-3 expression by Peripheral Blood Mononuclear Cells (PBMCs) obtained...

Cytotoxic effect of pyocyanin on human pancreatic cancer cell line (Panc-1)

Objective(s): Pyocyanin, a blue-green pigment produced by Pseudomonas aeruginosa, interferes with host redox cycles, which can lead to generation of reactive oxygen species and progressive cellular oxidative damage. The aim of this study was to assess the influence of pyocyanin on human pancreatic cancer cell line.Materials and Methods: Polymerase Chain Reaction (PCR) was applied to confirm the...

Altered Suppressor Function of Regulatory T Cells in Type 1 Diabetes

Background: Type 1 diabetes (T1D) is a T cell mediated autoimmune disease targeting the insulin-producing β cells within pancreatic islets. Autoimmune diseases may develop as a consequence of altered balance between regulatory (Tregs) and autoreactive T cells. Objectives: To evaluate Treg cells frequency and suppressive function in the peripheral blood of newly diagnosed T1D patients in compari...

Expression of TAAs in glioblastoma and expansion of anti-TAA -reactive T cells

Methods Fresh blood samples were obtained from 50 patients with tumors of the central nervous system and tested for antiTAA reactivity. T cells were expanded without cytokines, with IL-2 and IL-7, or with IL-2, IL-15 and IL-21 and tested for CD4/8 expansion by flow cytometry and for IFN-gamma production by ELISA. PBMCs were expanded using IL-2/15/21 and peptides (15mers) covering TAAs (survivin...
